Rapid Multiplication of Carnation through Somatic Embryogenesis from Flower Bud - induced Callus


Abstract
Experiments were conducted to develop methods of somatic embryo induction and maturation from embryogenic calluses of carnation, their germination in vitro, and their growth in greenhouse. The embryogenic calluses, induced from flower buds, had proliferated over 120 weeks without loss of embryogenic competence through periodic subculture with MS medium supplemented with 0.5 §·¡¤L^(-1) 2,4-D. When liquid agitation-cultured embryogenic calluses were transferred on to a regeneration medium (MS medium without 2,4-D, 3% sucrose, 0.5% Phyta gel), numerous i§®ature embryos were formed on the periphery of the calluses. The i§®ature embryos were matured with high efficiency on the above medium added with 7.5% PEG and 0.03 §·¡¤L^(-1) ABA. Hand-selected embryos were germinated in vitro on sponge mini plugs (12¡¿12¡¿15 §®). The plug plants which grown ca. 2 §¯ in height were transplanted into soil mixture of 72-cell plug trays, and grown to 10 §¯ seedlings within 4 weeks.
